I have a degree in Physical Therapy (2000), and both Masters (2002) and Doctorate degree in Morphology (2005). I was a Post-doc fellow of the Foundation for Research Support of Rio de Janeiro State (FAPERJ) for eight months working at the Tissue Repair Laboratory (Rio de Janeiro State University – UERJ) and then worked as a Visiting Professor for two years in the same university. I joined the Institute of Biomedical Sciences, Federal University of Rio de Janeiro in September 2009. From my initiation as a graduate fellow in March 2001 to December 2011, I have developed uninterrupted teaching activities in Basic Science area (Anatomy and Histology) and also clinical and research orientation in Physical Therapy. I have applied grants for CNPq (The National Council for Scientific and Technological Development) and FAPERJ in the area 2.06.00.00-3 (morphology) and 4.08.00.00-8 (Physical and Occupational Therapy). My research activities directed by Prof. Dr. Luis Cristóvão de Moraes Sobrino Porto (supervisor) during as a graduate fellow at UERJ contributed to the establishment and formation of three well-defined research lines coordinated by me at the Institute Biomedical Sciences. They are:
1) Experimental lung inflammation
This line of investigation was the first to be developed by me. By then, I was a graduate student. In 2001 there were not many papers about cigarette smoke in Brazil focusing on basic science. Back then, there were only two groups working with cigarette smoke: one group in São Paulo (University of São Paulo) and another group at the Federal University of Rio de Janeiro coordinated by Prof. Vera Lúcia Gonçalves Koatz from Institute of Medical Biochemistry.
In my opinion, the biggest issue regarding cigarette smoke studies is the time spent on exposure (the experimental model is chronic), which demands a daily exposition protocol (inhalation) that goes on during the weekends. Also, cigarettes are things not funded by the agencies, leading to a personal investment in this line of investigation. The biggest challenge was to create a chronic model with the shortest time possible. The literature reports a minimum exposure time of six months; however, six months is sufficient time for alveolar changes in non-exposed mice. After a few failed attempts, I found an exposure model of 60 days that caused emphysema, histologically similar to the experimental models of six months. This model is a success, yielded several publications and this model has been used by other groups in Brazil and even abroad. In fact, in Brazil, the cigarette smoke group in which I stand as a Principal Investigator is the most cited and it has the largest numbers of manuscript regarding cigarette smoke (24 papers in PubMed).
2) Oxidative stress
This research line was created focusing on studing specific markers of inflammation in experimental models developed in the laboratory and in which the primary stimulus is oxidative. Cigarette smoke itself contains many oxidants. Furthermore, emphysema, lung fibrosis and various acute or chronic lung lesions are based on pathophysiological imbalance between oxidants (generated by stimulation and inflammatory cells) and antioxidants (Vitamins, glutathione, superoxide dismutase, catalase, etc.) favoring oxidants. The study, characterization and development of oxidative stress methodologies opened doors for the study of other organs such as heart, liver, kidneys and central nervous system. However, these new approaches are still under implementation.
3) Extracellular matrix
This line of investigation was created in order to quantify and characterize non-fibrous and fibrous elements in target organs for the study from the experimental models described above. In general, the processes of tissue injury, tissue repair and remodeling occur in preference to a renewal of the elements from extracellular matrix. The special interest is collagen and elastic system. The amorphous substance is also to be studied in line of investigation, but the study and characterization are still under implementation.
