Retinal ganglion cells (RGCs) are the main output neurons that wire the retina with higher centers of the brain for the perception of vision. Degeneration of these cells in glaucoma patients disrupts this wiring leading to irreversible loss. Ex-vivo stem cell therapy for glaucomatous degeneration is a practical approach whose success depends upon transplanted human RGCs’ ability to elaborate axons that can read guidance cues and connect properly with the central targets. However, whether or not the hRGCs recruit similar mechanisms to find the central targets is not well understood. Here, we have examined if hRGCs are capable of reading the cues within the retina to orient their axons toward the optic disc for a proper exit, the first step toward connecting with the central targets.
