Real-time Quantitative PCR Detection of HPV mRNA and Its Relation to Cervical Cancer

Objective To establish a real-time quantitative reverse transcriptionPCR for detecting HPV mRNA expression,and to study the relationship between the quantity of HPV mRNA expression and the development of cervical cancer. Methods A total of 155 cervical biopsy specimens were taken for pathological diagnosis,at the same time,HPV DNA was detected by real-time quantitative PCR and HPV E7 mRNA expression was determined by real-time quantitative reverse transcription-PCR. Results Among all HPV DNA positive samples,mRNA expression in cervicitis and CIN I period specimens was both 0,while those in CINⅡ period specimens,CIN Ⅲ period specimens,and cervical cancer specimens were 50%,50%,and 87.5%,respectively.The expression levels increased in turn. Conclusions The expression rate and expression level of HPV mRNA are increased with the development of cervical cancer,and moreover,its expression level is positively correlated with the development of cervical cancer.The establishment of HPV mRNA quantitative detection method can provide an important clinical guidance for prevention,development monitoring and postoperative prognosis of cervical cancer.
