Study on the Interaction Between Shikonin and DNA by Fluorescence Spectroscopy

Berberine hydrochloride(BR)was utilized as a fiuorescence probe to study the interaction of shikonin(SHI)with herring sperm DNA in Tris-HCl buffer solution(pH7.4).The results showed that SHI competed with BR to bind with the site of DNA.The fluorescene intensity of DNA-BR was quenched by SHI via a mixture of static quenching and dynamic quenching mechanism.SHI interacted with DNA by intercalation and electrostatic gravity.Furthermore,the thermal denaturation experiment confirmed that intercalative binding was the main mode of interaction between SHI and DNA.At last,the calculated binding constant was 3.48×104 L·mol-1 by scatchard equations.
