Strategies for developing Nogo antagonists

Ian Ragan—Lilly Research Centre, Windlesham, UK This review is a very succinct account of ideas and developments in blocking Nogo action as an approach to the regeneration of axons in neuronal and spinal cord injury and autoimmune disease, such as MS. The GSK group have made important contributions to the area and discuss several strategies ranging from passive and active immunisation to antisense and blockade of the Nogo signalling cascade. The pros and cons of the various ideas are compared and the outstanding issues presented in what is now an exciting and fast-moving field with tremendous potential for patient benefit.
