To study features and functions of donkey Growth hormone gene. A pair of specific PCR primers was designed for cloning the coding sequence of GH gene from liver tissue. The fragment with 706 bp was got by RT-PCR amplifying, and analysis show it include a complete open reading frame, encoded 216 amino acids. Compared amino acids sequence with other animals, the sequences of donkey GH gene share 87.7%, 88.3%, 88.3%, 98.9%, 91.0%, 90.6%, 92.6%, 93.7%, 91.0% and 92.8% similarity with Guinea pigs, rats, mice, horses, goats, sheep, dogs, pigs, cattle and cats respectively, The protein to be codeing by cDNA of donkey GH gene had seven hydrophobic region, existence of seven transmembrane region and a signal peptide, is a kinds of secreted protein, the second structure main has a-helix and irregular curly and three-dimensional solution structure composed of 27 similar to 215 amino acids. It was very conservative of donkey GH gene in evolution, the phylogenetic tree constructed basing on CDS sequence is consistent with the results of comparative morphology and comparative physiology.
