•LdlrLec1 and LdlrLec2 expression were obviously upregulated upon WSSV challenge.•Injection of rLdlrLec1 and rLdlrLec2 into shrimp inhibited WSSV replication.•Knocking down the expression of LdlrLec1 and 2 increased WSSV replication in vivo.•LdlrLec1 and LdlrLec2 interact with VP28 of WSSV and inhibit the viral infection.
