4,7-Bis-chorosulfophenyl-1, 10-phenanthroline-2,9-dicarboxylic Acid (BCPDA)-europium labeling anti-alpha-foetoprotein (AFP) is described on the synthetic bifunctional BCPDA basis. B-CPDA can be incorporated into anti-AFP under relatively mild-conditions and complexed with Eu3+ form a complex of relative highly fluorescence intensity and bioactivity. A conclusion is obtained that the best, labeling condition is pH 9.1 (0.1 mol/L) carbonate, the dosage of BCPDA is 120similar to160 molar over-the anti-APP, reaction time is 50 min. At the same time, the intensity of fluorescence spectrum and bioactivity of Eu3+-BCPDA-anti-AFP are discussed. The best incubating time is 60 min, pH. is 7.8 in Tris-HCl butter solution. The detection limit for the determination BCPDA is 4.3x10(-11) mol/L when Eu3+ concentration is 10(-6) mol/L.
