Microchip Capillary Electrophoresis For Determination Of Small And Dense Low-Density Lipoprotein Assay

In this study, the separation of serum small, dense low-density lipoproteins ( sdLDL ) was carried out by microchip capillary electrophoresis. The optimun experimental conditions were 40 mmol/L Tricine, 40 mmol/L N-methyl-D-glucamine buffer ( 0. 1 mmol/L SDS was chosen in the sample solution and the running buffer contained 0. 02 mmol/L SDS). The specificity and saturation of 6-( ( N-( 7-nitrobenz-8-oxa- 1, 3-diazol-4-yl) amino) hexanol) sphingosine ( NBD ) C-6-ceramide as lipoprotein dyes, the effect of assay time and intermediate-term storage of serum samples on electrophoresis behavior were studied. The roles of sodium dodecyl sulfate ( SDS ) to minimize lipoprotein adsorption and improve the resolution of lipoprotein separation were also investigated. The proportion of sdLDL ( 75. 0% ) in patients with coronary heart disease ( CHD ) was significantly higher than that in healthy controls ( 6. 0%, P < 0. 01). These results suggested that the microchip-based assay was a simple, rapid and highly efficient technique and could be used as routine analysis to evaluate the CHD risk.
