•CFMA allows direct observation of single cell mast cell degranulation in real time.•Both CXCL10 and RANTES, airway smooth muscle-relevant cytokines, are capable of directly inducing mast cell degranulation.•Mast cells are capable of stimulation pathway-dependent regulation of exocytosis.
