•Preclinical studies show insulin-like growth factor 1 receptor (IGF-1R) and poly-ADP-ribose-polymerase (PARP) to be targets for Ewing sarcoma.•It is very important to understand the biological rationale for patient selection.•Therapeutic strategies with IGF-1R or PARP should definitely be explored further.
