•Ambulatory measures of joint mechanics are associated with serum COMP increase.•Ambulatory mechanics explain some (61%) of the variance in serum COMP change.•Ambulation at greater speeds results in a greater serum COMP concentration increase.•Serum COMP returns to normal levels within 30min after ambulating 4000 steps.
