reversed these effects (p,0.001).As a functional proof of the DARPP-32-SRP-20 axis on splicing, we tested the expression of CD44.Our results demonstrated that DARPP-32 regulated the SRP-20 dependent CD44 v8-v10 (CD44E) splicing in gastric cancer cells.Using oligos that span different exons of CD44, qRT-PCR and direct sequencing, we have confirmed the specific regulation of CD44E by DARPP-32.We have also examined the function of CD44E and discovered that it plays an integral role in promoting AGS cancer cell invasive activity (p,0.001).Conclusion: Our results demonstrate, for the first time, the role of DARPP-32 in regulating the splicing factors in cancer cells.The DARPP-32-SRP-20 axis played an important role in regulating the CD44E splice variant, which also played a crucial role in promoting gastric cancer cell invasive activity.This novel splicing-dependent mechanism provides a new paradigm in the promotion of invasion and metastasis in cancer cells.
