•Identified the first major Cry2Ab2 resistance allele in a field collected Diatraea saccharalis population.•Resistance to Cry2Ab2 in D. saccharalis was inherited as a single autosomal gene.•The Cry2Ab2 resistance in D. saccharalis was non-recessive.•No fitness costs were associated with the Cry2Ab2 resistance in D. saccharalis.
