•A theoretical analysis of the incomplete depletion of fluorescence in STED-FCS process.•A numerical analysis of the STED effective fluorescent volume.•The performances of different typical fitting models are compared.•A STED-background corrected fitting model is proposed to retrieve unbiased diffusion times and average molecule numbers.•A validation of the new fitting model on experimental STED-FCS measurements is provided.
