•AVT potently stimulated ACTH release from the bullfrog pituitary.•Homologous CRF stimulated ACTH release very moderately.•CRF acted synergistically with AVT, enhancing ACTH release markedly.•AVT V1b-type receptor mRNA was abundantly expressed in the ACTH cells.•AVT is the main ACTH-releasing factor acting via V1b-type receptor.
