•Higher circulating IL-6 is associated with short sleep duration in midlife adults.•Short sleep on IL-6 levels is moderated by variation in the GR gene (NR3C1).•CRP levels are elevated in association with both short and long sleep duration.•The effect of sleep on CRP levels was not moderated by NR3C1 variation.
