e22018 Background: CTCs can be detected in the blood of men with prostate cancer and have been used as a biomarker. The only FDA-approved technology (CellSearch) utilizes epithelial cell adhesion molecule (EpCAM) expression for CTC detection. Methods: We aimed to detect CTCs in the blood of patients with mCRPCa using both CellSearch and a novel microfilter-based device. Detection by CellSearch was performed according to standard protocol. For the microfilter-based capture peripheral blood from mCRPCa patients was diluted 1:1 with PBS and 1% formalin. Cells were captured with microfilters and subsequently stained with pancytokeratin (CK) and CD45. CTCs were identified as large CK+/CD45- events with morphologic criteria consistent with a tumor cell. Data were analyzed using non-parametric methods: Spearman rank correlations, Mann-Whitney test, and the Jonckheere-Terpstra test.
