•A new class of sulfonyl hydrazones was discovered as MAOs inhibitors.•All compounds had IC50 values in lower micro-molar range.•Mode of inhibition was found in enzyme kinetics studies.•A new binding pocket for non-competitive MAO-A inhibitors was discovered.•All compounds have highly favorable ADME profile with good oral bioavailability.
