•In early S-phase arrested cells by HU essentially all rRNA genes have an open chromatin.•Only a portion of all open rRNA genes has RNA polymerases I.•rRNA genes remain poised for transcription in cell cycle arrested cells.•rRNA processing and synthesis are compromised in HU treated cells.
