•Single-molecule FRET of cytosolic CRAF were successfully measured in live cells.•CRAF has at least three conformations, the closed, open and fully-open form.•EGF stimulation conforms a small fraction of wildtype-CRAF to the fully-open form.•S612A mutant CRAF adopts the fully-open structure.•State transitions between the closed and open conformations were detected.
