Production of Monoclonal Antibody Specific to Campylobacter jejuni

Campylobacter jejuni is an important cause of gastroenteritis in human. Immunological tests are needed for accurate and rapid identification of Campylobacter. This study aimed to produce monoclonal antibodies against C. jejuni isolated from Thai patients. BALB/c mice were immunized with either whole cell homogenate antigens or lipooligosaccharide of Campylobacter jejuni 43. The immunized spleen cells were fused with P3X myeloma cells. Thirteen hybridomas were produced. The hybridoma clones were selected using indirect ELISA and western blot analysis. Hybridoma clone 2D10 reacted with C. jejuni 43 but not with E. coli K12 by indirect ELISA. By Western blot analysis, antibody from clone 2D10 bound to proteins migrating at molecular masses ranging from 17-150 kDa. Clone 2D10 was selected to re-clone and expanded by limited dilution technique and kept in liquid nitrogen for further characterization.
