•Prenatal exposure to ethanol affects the structure and function of the dentate gyrus of adolescent macaques.•Ethanol targets postnatal neurogenesis, notably proliferating cells and those exiting from the cell cycle.•The structural effects correlate with ethanol-altered behavior, i.e., performance in a visual-paired comparison task.•The time of gastrulation is a critical window of vulnerability.•Gestational exposure to ethanol affects long-term fetal programming of cognitive dysfunction.
