•Tissue plasminogen activator (tPA) has immunomodulatory effect in EAE.•Absence of tPA aggravate EAE, while administration of tPA ameliorate the disease.•tPA−/− mice show decreased T-cell reactivity and reduction in T-regulatory cells.
