•Regulation of Th17/Treg cells by a combination of L. acidophilus, vitamin B, and curcumin via downregulation of pSTAT3.•Suppression of IL-17 and induction of IL-10 by the combination of L. acidophilus, vitamin B, and curcumin in human PBMCs.•Suppression of osteoclastogenesis by the combination of L. acidophilus, vitamin B, and curcumin in murine monocyte•The combination of L. acidophilus, vitamin B, and curcumin decreased pain and pro-inflammatory cytokines in OA animal model.•Regulation of anabolic and catabolic activities by the combination of L. acidophilus, vitamin B and curcumin in vitro and in vivo.
