RAPD ANALYSIS OF BULGARIAN PEPPER INDUCED MUTANTS

Induced mutagenesis is among the key methods used by modern breeding. The successful application requires characterization of the induced changes. In this work we have analyzed seven pepper genotypes produced by induced mutagenesis. These genotypes carry on different traits like high beta-carotene content, male sterility, early ripening, and lack of anthocyanin. The lines were analyzed by RAPD using 40 decamer primers (Carl Roth) in order to identify DNA patterns useful for breeding purposes. We have found 16 primers generating profiles suitable for genotype identification. All selected primers have generated clear patterns containing 5 to 16 well defined bands ranging from 300 to 3000 bp. Primers A13, A18 and B01 generate polymorphic bands in genotype 1928. Primer B10 generates polymorphic bands in genotypes 1928 and 1966. The polymorphism percentage between lines of interest and the rest of the screened material ranged from 7.69% to 21.43%. As a result we have found primers for differentiating genotypes 1928 and 1966 from the rest of the breeding material.
