Mesenchymal stem cells inhibit T-lymphocyte proliferation through secreting TGFbeta

Objective:To study if MSCs inhibit T-lymphocyte proliferation through secretion of transforming growth factor-beta (TGF-β1).Methods:The level of TGF-β1 was evaluated by enzyme linked immunosorbant assay (ELISA);after the TGF-β1 was neutralized by its correspondig mAb,the ability of T lymphocyte-proliferation was measured by ~~ 3H -Thydimine incorporation;and the level of interferon-gamma (IFN-γ) was measured by Eli-spot kit.Results:MSCs reduced the level of IFN-γ secreted by activated T lymphocytes;inhibited the ability of T-lymphocyte proliferation and such an effect could be reversed partly by anti-TGF-β1 antibody.Conclusion:MSCs inhibit T-lymphocyte proliferation through secreting TGF-β1.
