•Climacteric fruits accumulate a variety of 5-CH3-THF polyglutamylated profiles.•Postharvest ripening increased 5-CH3-THF with short Glun chains in all fruits tested.•Ethylene treatment only decreased 5-CH3-THF with long Glun chains in ripe fruit.•Plant FPGS and GGH genes have evolved separately within the plant kingdom.•Papaya FPGS SF6 has a unique primary sequence along with the very-long Glun profile.
