•The vitamin D active metabolite calcitriol plays protective roles in the prostate.•Intraprostatic levels of calcitriol decrease with advancing age.•Expression of the enzymes CYP27B1 and CYP24A1 was linked to calcitriol levels.•Enzymatic and hormonal changes were in parallel with the onset of prostate lesions.
