花楸猕猴桃根对大鼠肝细胞CYP1A1酶的抑制作用

Abstract
目的:探讨花楸猕猴桃根对肝细胞CYP1A1酶的作用机制。方法:40正常SD大鼠随机分为空白对照组和猕猴桃根提取物高、中、低剂量组(50 g/kg、25 g/kg、13 g/kg),连续灌胃7 d,2次/d,处死大鼠后取出肝脏,通过检测N-脱甲基酶活性反映细胞CYP1A1酶活性,提取组织RNA逆转录后用RT-PCR检测CYP1A1 mRNA表达水平。结果:与空白对照组相比,花楸猕猴桃根高、中、低剂量组CYP1A1酶活性均明显受到抑制,且呈浓度依赖性( P﹤0.05),但各剂量组间无显著性差异;与空白对照组比较,花楸猕猴桃根高、中、低药物剂量组能显著抑制CYP1A1 mRNA的表达( P﹤0.05)。结论:花楸猕猴桃根可下调大鼠肝细胞CYP1A1酶的活性,并抑制其mRNA表达水平,这可能是花楸猕猴桃根能够预防癌症的作用机制。
