•Budding yeast Eco1 interacts with replisome via Mcm2 as well as PCNA.•Mechanism of Eco1-MCM interaction is different from that reported in human.•Disruption of Eco1-replisome interaction results in Eco1 protein destabilization.•Eco1 stabilization by replisome may control proper cohesion formation.
