The invention discloses a method of in-vitro expression of recombinant pectin methylesterase PbrPME and coding genes and application of the recombinant pectin methylesterase PbrPME. The method comprises the steps: adopting a designed primer to clone pear PbrPME genes, constructing a recombinant expression vector PbrPME-pCold-TF for Escherichia coli, transforming the constructed recombinant expression vector PbrPME-pCold-TF in Escherichia coli Rosetta (DE3), constructing a recombinant strain for expression of the pear PbrPME protein, expressing the recombinant strain, purifying the recombinantpear PbrPME protein through nickel-column affinity chromatography, treating a pear pollen tube by using the recombinant pear PbrPME protein, counting the length of the treated pollen tube, and verifying the activity of the recombinant pear PbrPME protein. Through the method, in-vitro expression and application of the pear PbrPME protein are realized, and requirements of related experiments can bemet fully.
