•Determination of resolvins in biological matrices by means of HPLC-MS/MS.•Combination of LLE and μSPE for extraction and clean-up.•Effective clean-up with ion suppression below 20 % for the different matrices.•Method validation on in 3 different matrices (cells, plasma and exudate).•Sample enrichment for high sensitivity with LOQ between 1−38 pg mL−1.
