•Pretreatment with IGF-I increased leucine's action on mTORC1 in muscle cell lines.•IGF-I increased Sestrin2 mRNA and protein levels in muscle cell lines.•Sestrin2 was involved in the action of leucine and IGF-I on mTORC1.•Increased Sestrin2 may play a role in enhancing the effect of leucine on mTORC1.
