•Neuroinflammation is a common feature of age-related brain disorders including AD, PD and cerebral ischemia.•The proinflammatory enzyme 5-LOX is a novel vital player in the development of age-related brain pathology.•Inhibition of 5-LOX activation exerts antiinflammatory and neuroprotective effects.•5-LOX acts as an emerging therapeutic target for age-related brain disorders.
