Effects Of Different Image Acquisition Modes On Analyzing Dna Degradation Of Human Splenic Lymphocytes

To evaluate the stability of CIAT on deducing the PMI, we quantitatively analyzed DNA degradation status of human splenic lymphocytes by using the same image analysis system and different image acquisition modes.. Methods: Selected 21 cadavers whose accurate PMI were known, sampled the spleen tissue and smeared the samples respectively every hour from the 5th to 36th hour after death. The smears stained by Feulgen-van's method, collected images by different image acquisition modes (HAPIS-1000 and Motic Med 6.0 CMIAS) and analyzed these images by the same image analysis system (Motic Med 6.0 CMIAS). Chose the geometric parameters (A, ED, ID) and grayscale parameters (AG, AOD, IOD) as detect indexes, took the mean of the data of 3150 cells as each statistic. Compared the differences between the two groups of measured means of the same kind parameter under the different image acquisition modes. Results: No differences found between the corresponding same geometrical parameters with different image acquisition modes had statistic significances, but the differences of absolute means of same grayscale parameters had statistic significances, converted the means of grayscale parameters into percentages, no statistic differences were found. It suggests the two image acquisition modes have the same intrinsic basic rules, both can reflect the regularity of DNA degradation status effectively after proper conversion and calibration.
