Synthesis And Physicochemical Mechanistic Evaluation Of Chitosan-Based Interbiopolyelectrolyte Complexes For Effective Encapsulation Of Olz For Potential Application In Nano-Psychiatry

The design and development of more effective biopolymer-based drug delivery and controlled release systems are required to improve pharmaceutical products for psychiatric terapies. In this paper, the synthesis of three types of chitosan-based interbiopolyelectrolyte complexes (IBPECs) with olanzapine (OLZ) were evaluated. The proposal is an innovative ecofriendly formulations with potential application in nano-psychiatry. IBPECs were strategically synthesized following zeta potential measurements using chitosan (Ch) as the base material and tripropylphosphate (TPP) and alginate (ALG) as cross-linking biopolymers. The effect of TPP and ALG as a crosslinking agent on particle size, surface charge density and colloidal stability in complex formation of TPP/Ch, ALG/Ch and ALG/TPP/Ch were evaluated before and after the incorporation of OLZ at pH = 5.0. The zeta = f (pH) and zeta = f ([TPP/Ch], [ALG/Ch] or [ALG/TPP + Ch]) profiles allowed us to elucidate the supramolecular assembly mechanisms of each formulation and determine the best physicochemical conditions that maximize OLZ encapsulation.
