•The active ingredients in peanut skin against CIT was firstly clarified.•A facile method of HSCCC with pre-HPLC was developed to isolate PS-1–4.•PS-1 promoted the proliferation and maturation of MKs by JAK2/STAT3 pathway.•PS-1 was the first reported naturally-derived small molecule to treat CIT.
