•Genetic changes of primate and murine NXT1 and NXT2 were characterized.•NXT1 has been subject to functional constraint in both phylogenic lineages.•NXT2 has undergone different evolutionary fates in primates and murines.•The murine NXT2 has evolved conservatively.•In primates, NXT2 exhibits clear sign of adaptive diversity.
