Efficient Development of Antibacterial (−) -Epigallocatechin Gallate-Pbca Nanoparticles Using Ethyl Acetate As Oil Phase Through Interfacial Polymerization

Spherical (-)-epigallocatechin gallate (EGCG)-poly(n-butyl cyanoacrylate) (PBCA) nanoparticles (NP) with smooth surface were prepared through interfacial polymerization using ethyl acetate (EtOAc) as the oil phase solvent. The prepared EGCG-PBCA NP-water and EGCG-PBCA NP-oil showed high stability and dispersity observed by dynamic laser scattering (DLS). The structural analysis by X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) revealed that the interaction forces between EGCG and PBCA might be contributed mainly by hydrogen bonds and electrostatic interactions, which was also supported by molecular docking simulation. The encapsulation efficiencies of EGCG in EGCG-PBCA NP-water and EGCG-PBCA NP-oil were 76.30 +/- 1.81% and 74.60 +/- 2.32%, respectively. After encapsulation, properties of EGCG were improved, including sustained release, strong antioxidant activity, and enhanced antibacterial activity. The present study provides an efficient approach to encapsulate EGCG by PBCA using EtOAc as the oil phase which needs not pH adjusting of the water phase.
