Effects of Morroniside on Local Axon Regeneration and functional Recovery After Spinal Cord Injury in Rats via the Eph/ephrin B3 Signaling Pathway

This study was designed to investigate the effects of morroniside on local axonal regeneration and functional recovery after spinal cord injury (SCI) in rats via the Eph/ephrin B3 signaling pathway. Thirty SD healthy male rats, 10 assigned to the sham group, and another 20, were used to establish the spinal cord injury model, and the rats in the sham and model groups were treated by intragastric administration of normal saline, and the rats in the morroniside group were treated by intragastric administration of morroniside (60 mg/kg) using inflammatory factors [TNF-alpha,IL-6,IL-10], and oxidative stress factors Sub > [H2O2, MDA, 8-OHdG], Eph, ephrinB3expression, local axonal regeneration, and functional recovery. The BBB scores ofthe model group were low at 1D, 3D, 5D, and 1W after successful modeling and did not differ much over time (p < 0.05), the BBB scores of the model rats increased with time (p < 0.05), the levels of TNF-alpha, IL-6, IL-10, H2O2, MDA, 8-OHdG increased abnormally in the model group compared with the sham group (p < 0.05),and the BBB scores increased with time (p > 0.05) The levels of TNF-alpha, IL-6, IL-10, H2O2, MDA, and 8-OHdG were decreased (p > 0.05) in the model group, Eph and ephrin B3 were abnormally increased in the model group compared with the sham group, and Eph and ephrin were increased in the model group compared with the sham groupB3 expression was decreased (p < 0.05). The surface area positive for NF-200, synaptophysin was higher in the model groupcompared to sham group, and the surface area positive for NF-200, synaptophysin was lower (p < 0.05) in morroniside group compared to model group. The intervention of SCI model rats with morroniside, which is able to modulate the Eph/ephrin B3 signaling pathway and promote the local axonal regeneration situation as well as the recovery of function in rats, is of great clinical value for promotion.
