•CAR-T cell therapies exhibit distinct cellular kinetic profiles that influence clinical outcomes.•CAR-T cellular kinetics are less favorable in solid tumor settings.•Mathematical modeling methods are commonly applied to assess, characterize, and predict the cellular kinetics of CAR-T cells and their relationship to tumor killing.•The clonal heterogeneity of CAR-T cell therapies may confound direct linking of cellular kinetics to efficacy.
