•The nitrogen and phosphorus removal in an anammox reactor was achieved by Fe2+.•0.12 mM was the optimal dosage of Fe2+ to strengthen simultaneous N and P removal.•T-EPS was a major site for biomineralization due to its better adsorption properties.•Fe2+ weakened the competition for substrates between anammox and denitrification.•The gene abundances related to electron behavior were enhanced by Fe2+.
