•Rationally designed thiazolidine-2,4-dione based analogs were synthesized.•Structure activity relationships were explored.•A number of compounds exhibited higher potency than reference drug safinamide.•Compounds showed BBB penetration in PAMPA in-vitro assay.•Non-neurotoxic and reversible.
