Abstract
目的:比较吸入氩气(inhaled argon,iAr)和吸入氢气(inhaled hydrogen,iH2)对小鼠急性脑缺血再灌注(cere-bral ischemia/reperfusion,CIR)损伤后神经功能改善的作用.方法:选取48只健康成年SPF级雄性ICR小鼠,将其随机分为假手术组(Sham组)、局灶性大脑中动脉栓塞(transient middle cerebral artery occlusion,tMCAO)模型组(tMCAO组)、iAr治疗组(tMCAO-iAr组)、iH2治疗组(tMCAO-iH2组).于再灌注24 h后行神经功能评分,采用2,3,5-氯化三苯基四氮唑(2,3,5-triphenyltetrazolium chloride,TTC)染色观察急性脑梗死损伤程度,同时监测术后体质量和存活率的变化,评估两种气体的神经保护疗效差异.结果:与tMCAO组相比,iAr和iH2治疗均可明显改善神经功能(P<0.01),脑梗死体积、体质量损失均明显减少(均P<0.001),存活率明显提高;组间相比,tMCAO-iAr组在神经功能评分(P<0.01)和皮层梗死体积两个指标上更优于tMCAO-iH2组.结论:再灌注后尽早iAr和iH2对CIR损伤都能起到一定的保护作用,且iAr对神经功能改善和皮层脑区的保护作用更好.
