•vYF is a new developemental live-attenuated YF vaccine based on a strain cloned from YF-VAX® virus strain.•vYF virus strain is adapted for growth in Vero cells cultured in serum-free media.•vYF has a satisfactory safety profile and elicits a robust immune response in macaques.•vYF provides effective protection against challenge with wild-type YFV Asibi.
