•Polymorphic alleles of either ABCB1 or VEGF genes predicted better outcome in patients with MCL receiving LEN after ASCT.•ABCB1, NCF4, GSTP1, and CRBN polymorphisms were associated with toxicity during induction and with LEN dose reductions.
