A, Rh30 cells were exposed to AZD8055 (2 M) for the times indicated and total proteins were extracted for immunoblotting to detect FANCD2. B, Rh30 cells were transfected with vector, AKT1-Myc plasmid encoding wild type AKT1. 48 hr later, total proteins were extracted for detection of FANCD2, GSK3, pGSK3-S9, AKT and pAKT-S473. Myc antibody was used to show the expression of AKT1 wt with Myc tag. C, Rh30 cells were transfected with vector, Myr-AKT1 plasmid encoding constitutively active AKT1. 48 hr later, total proteins were extracted for detection of FANCD2, GSK3, pGSK3-S9, AKT and pAKT-S473. D, Rh30 cells were treated with rapamycin (100 ng/mL), AZD8055 (2 M) or MK2206 (10 M) for 16 hr. Total proteins were extracted for immunoblotting of FANCD2, S6K1, pS6K1-T389, AKT and pAKT-S473. GAPDH and -Actin served as loading control
