•LncRNA MSTRG91634.7 could modulate the silica-induced epithelial injury through targeting PINK1.•Exosomal lncRNA MSTRG91634.7 derived from macrophage restrict silica-stimulated EMT by restoring mitochondrial homeostasis of epithelial cell.•LncRNA MSTRG91634.7/PINK1 regulatory axis could attenuate silica-induced inflammation and fibrosis through modulating EMT and mitochondrial homeostasis in mice lung.
