•High pressure freezing and cryo-sectioning of protein crystals can be used for 3D electron diffraction studies.•Complete diffraction data at 2.9 Å resolution have been obtained from hen egg white lysozyme crystal cryo-sections.•High pressure freezing and cryo-sectioning preserve macromolecular structures at quasi-atomic resolution.
